MtoZ Biolabs Developed Acetyl-proteomics Analysis Platform

Acetylation is a highly conserved, reversible protein modification in vivo that plays an important role in the activation of transcriptional regulators in the nucleus. In addition, a large number of non-histone acetylation modifications are involved in the regulation of metabolic pathways and metabolic enzyme activities.

Due to the low content and wide dynamic range of acetylated proteins in biological samples, the acetylated peptides need to be enriched to improve their abundance before mass spectrometry, and then the enriched acetylated groups are obtained by traditional quantitative proteomics analysis. The peptide samples are subjected to quantitative analysis. MtoZ Biolabs uses peptide pre-separation to reduce the effect of high-abundance histones on protein acetylation identification, and then combines immunoprecipitation with high-efficiency antibody-enriched acetylated peptides to identify and quantify large-scale acetylation. The selected kit is an internationally accepted acetylated peptide-specific enrichment kit supplied by CST. In order to identify and quantify more acetylated peptides, they use 2-3 different enzymes to digest the protein samples during the digestion process, ensuring that the peptides with acetylation sites can entered into the mass spectrometer for analysis with the way of high-efficiency ionization to eliminate the loss of acetylation information due to the inappropriate peptide length and low ionization efficiency.

Thermo Fisher's Q ExactiveHF mass spectrometry platform, Orbitrap Fusion mass spectrometry platform, Orbitrap Fusion Lumos mass spectrometry platform and Nano-LC are used by MtoZ Biolabs to launch the acetylated quantitative proteome analysis service.
